Abstract: We optimized the skinned fiber technique and high resolution respirometry developed for the examination of mitochondrial function. Storage of skeletal muscle fibers in ice-cooled storage buffer for 36 hours did not caused a significant change in mitochondrial function as determined by respiration of complex I and II dependent substrates. However, stable mitochondrial function was only guaranteed if cooling was without interruption. The newly established multiple substrate-inhibitor titration provides the possibility to measure complex I dependent functional deficits. This parameter was shown to be independent of the mitochondrial content of skeletal muscle fibers. Furthermore, we showed that shipment of samples in ice-cooled storage buffer by mail did not significantly influence mitochondrial function measured by respirometry.
