Several in vitro models have been developed for studying skeletal muscle development including synaptogenesis of the neuromuscular junction and skeletal muscle regeneration. In vitro skeletal muscle can be studying on models of intact isolated myofibers, cultured myoblasts derived from muscle tissues or immortalized muscle cell lines. Co-cultures of rat embryonic spinal cord explants with human muscle cells are most advanced system used in studies on the synaptogenesis of neuromuscular junction, since this system offers significant advantages and enables to study functional neuromuscular junction which result in muscle contraction. Namely human muscle does not spontaneously contract unless it is innervated or stimulated with electric pulses. Myoblasts derived from satellite cells when placed in appropriate in vitro conditions (which can be modified for different physiological and pharmacological studies) proliferate and at certain density start to fuse and form multinucleated myotubes. In case of experiments on the innervated muscle fibers, an explant of the rat embryonic spinal cord is placed on a monolayer of muscle cells. In those co-cultures motor neurons functionally innervate myotubes which then start contracting. These co-cultures are long-lived and contraction can be maintained for several months. Since the process of human muscle regeneration in many respects resembles the process of myogenesis, stages of muscle regeneration including satellite cell activation, myoblast proliferation, fusion into myotubes and nevertheless innervation can be reproduced and follow in the experimental model of the in vitro human muscle. In vitro model can be used as an experimental model for signalling pathways, metabolic adaptation, mitochondria, cytokine secretion studies involved in contraction and regeneration process.
