Description

Go in DatLab to Mark statistics (F2), select which type of marks you want to export ("All marks in plot" or "DL-Protocol marks", with 3 possibilities each), then click on [Copy to clipboard] to copy selected values and paste them to a DatLab-Analysis template for numerical and graphical data analysis.

DatLab-Analysis templates for O2k-analysis of respiratory flux - strategy

DatLab-Analysis templates are made available for specific SUIT protocols, to express respiration as oxygen flux or oxygen flow normalized for sample concentration, and as flux control ratios, FCR and flux control efficiencies.

Guidelines

  1. Select the DatLab-Analysis template for your specific SUIT protocol. In DatLab, in the pull-down menu 'Protocols: SUIT: Browse DL-Protocos and templates', select your specific SUIT protocol and open the chosen DatLab-Analysis template.
  2. Make sure that the DatLab-Analysis template corresponds with the protocol of your experiment.
  3. Create a copy for your data analysis.
  4. In the copied Excel file, select the tab "Data".
  5. In DatLab go to Mark statistics (F2), select in "DL-Protocol marks": Slope uncorrected + all info. Click on [Copy to clipboard] to copy selected values.
  6. In the DatLab-Analysis file paste the selected values in the cell A7.

Concept

  1. The normalization for sample concentration is transferred into a DatLab-Analysis template, since (1) in some cases the final sample concentration is not known until a later time point (e.g. protein or CS determination), and different markers may be used for different aspects of the analysis; (2) the sample (isolated mitochondria, homogenate, cell suspensions) is diluted during titrations, and the dilution effect is calculated in the DatLab-Analysis template.
  2. A logical next step: instrumental O2 background parameters may not be known at the time of the experiment, particularly when brackting instrumental quality control is performed (before and after the experiment). Finally optimized background parameters may be calculated at the end of a complete experimental series, combining several relevant instrumental O2 background tests. Up to now, the DatLab-Analysis files do not document the instrumental O2 background parameters. For quality control each individual DatLab file has to be checked. Therefore, the logical next step is to export from Mark statistic only slopes (O2 slope neg.) for complete data analysis in the DatLab-Analysis template. There, the instrumental O2 background correction is easily implemented, adjustments of parameters are quickly performed, actual parameters used can be much more quickly checked, and documentation for tracing is provided in the DatLab-Analysis files in a more complete and compact form.

  • DatLab-Analysis templates are provided for DatLab 7. For using previous DatLab-Analysis template/own Excel templates:
  1. Select a Layout for DatLab graphs with the plot of O2 flux or O2 flow normalized for the sample concentration.
  2. Set marks on the plot for O2 flux or O2 flow.
  3. Press [F2] (or click on Mark statistics) and select "Flux/flow" marks. Click on [Copy to clipboard] to copy selected values.
