Work Plan summary
Aim and motivation
A collaboration already exist between the two partners involved in this STSM. This collaboration includes two main projects: the study of mitochondrial function in white adipose tissue (WAT) in order to understand its role and importance in the hallmark of type 2 diabetes; and the implication of both parts in the WG2 and WG3.
Proposed contribution
This STSM will be a first step into the assessment of most used protocol for the study of mitochondrial respiration in white adipose tissue preparations (not isolated mitochondria), given the lack of standardized protocols comparisons and the aims of the COST Action.
Techniques
Oroboros O2k will be used for measuring mitochondrial respiration in different type of white adipose tissue sample preparation (e.g. whole WAT homogenate, adipocyte after SVF removal, and permeabilized WAT depot after gentle centrifugation to remove lipid layer)
Planning
1 day for study design and clarifying all the variables involved for a proper experimental performance
4 days for experimental performance
1 day for data analysis
4 days for data discussion, along with WG2 and WG3 results discussion
