MitoEAGLE Short-Term Scientific Mission

Work Plan summary
I aim and motivation Obviously my aim is to understand mitochondrial physiology. I am member of scientific group concerned with hepatocytes, especially their metabolism. Measuring utilization of oxygen by hepatic mitochondria is a great way how to observe metabolic changes caused by different conditions, in my case NAFLD (non-alcoholic fatty liver disease) caused by high fat diet. This disease is wide spread at USA and Europe and the number of affected patients is increasing. The pathophysiology of NAFLD has not been sufficiently explained yet. I hope, that characterization of respiratory changes in NAFLD mitochondria will be supportive to elucidation and curing NAFLD.
As far as I am student of medicine and hopefully I will become a doctor, I find knowledge of mitochondrial physiology very beneficial for my further specialization. II. Proposed contribution As I mentioned, I am concerned in changes of mitochondrial respiration caused by NAFLD (non-alcoholic fatty liver disease), I would like to extend my knowledge about mitochondrial physiology. MitoEAGLE project is a great opportunity to get deeper into a topic and also get other points of views. III. Techniques The instrument I am interested in is an Oroboros instrument. I hope that during the meeting I will find further ways of usage and also its limitation. The most important I find discussion about interpretation of results obtained during measurements. Last but not least MitoEAGLE project is also focused on proper terminology usage, very important for understanding and sharing outcomes and theories. IV. Planning I would like to extend the project I am participating now. Our recent outcomes show, that long time high fat diet, leading to NAFLD, significantly decreases ETS capacity (electron transfer system capacity). Some studies shown, that some disturbances at respiratory chain might lead to mitochondrial proliferation. I would like to investigate this possibility and its further impact on development of NAFLD. According to my next carrier steps, I hope I will finish my medicine studies in two years and become a doctor specialized in diseases of metabolism. After finishing my MD degree I would like to continue my research as a Ph.D. program student.
