The default setting of Slope smoothing is 40 data points, corresponding to an interval of 80 s at a data recording interval of 2 s. DatLab 7 expresses it in data points and DatLab 8 in seconds.

Do I need to change the filter set of the O2k-Fluo Smart-Module?

Because two different filter sets can be used with the blue Fluo-Sensors, filters should be changed according to the fluorophore that will be used. Extra filters are provided for replacement in case of damage.

How often should I re-assemble the O2k-chamber?

We recommend disassembling, cleaning and re-assembling the O2k-chamber at least every 1-2 years. It is also necessary to disassemble it when there is persistent biological or protein contamination, medium leakage from the chamber, an oxygen leak of the chamber, or an unstable or unexpectedly high or low oxygen signal.

Noticing higher values of J°1 when using the O2k-Chamber sV (0.5 mL) compared to the 2 mL O2k-Chamber?

The instrumental oxygen background flux, J°O₂, is volume-dependent in the closed chamber; therefore, the values of J°1 are about four times higher in the sV chamber.

Can I compare data sets recorded with different sensor gain settings?

Oxygen signals obtained after proper calibration are independent of the gain settings, therefore oxygen concentrations (and fluxes) can be compared, as long as the corresponding air and zero calibrations are applied.

Add catalase when measuring Complex IV (CIV) activity in permeabilized fibers (pfi)

Pfi have a high content of iron-containing proteins, which accelerate the autoxidation of tetramethyl-p-phenylenediamine (TMPD) in the CIV assay. Catalase prevents the accumulation of H₂O₂ generated by this autoxidation.
