MiPNet Reference Laboratories

OROBOROS INSTRUMENTS integrates the concept of open innovation in terms of (i) our Open access approach to the O2k-Manuals, O2k-Protocols, O2k-Publications and open dissemination of methodological and scientific concepts in our O2k-Workshops and on the Bioblast wiki, and (ii) the open exchange of ideas and expertise through the WorldWide Mitochondrial Physiology Network of OROBOROS Reference Laboratories. For us, open innovation is an expression of Gentle Science.


OROBOROS

Our open innovation management is minimally bureaucratic, is not complete nor comprehensive, but attempts at acknowledging explicitly the outstanding and most valuable contributions and encouriging all OROBOROS O2k-users to participate and benefit from this concept on cooperation and feedback in science. Ideas that you communicate with us will be posted on this page. We will add comments and evaluations which may origin from the entire network, and will provide the links to results that emanate from the steps taken with the support by open innovation.

Hickey AJ: O2k-Fluo

NZ Auckland Hickey AJ - Tony Hickey was the first to implement optical sensors for O2k-Fluorometry through the window of the O2k-Chamber rather than through an additional port drilled through the stopper (Hickey 2012 J Comp Physiol B). This avoids complications of oxygen leakage and trapping gas bubbles in the chamber and minimizes optical disturbances originating from the rotating stirrer. This concept is now implemented in the O2k-Fluo LED2-Module. After going into series production, OROBOROS INSTRUMENTS sent a free O2k-Fluo LED2-Module to the lab of Tony Hickey. Tony Hickey is still involved with the project MitoCom, further developing the combination of high resolution respirometry and fluorometry. E.g. The initial contact to the company producing the LED currently most promising for measuring NADH fluorescence was via Tony Hickey. His group is also testing the O2k-Fluoreseceene module via alternative light sources for special applications.

Neufer PD: Measuring mitochondrial membrane potential in permeabilized fibers via the TPP method (and more)

  • Extending the TPP method to measure mitochondrial membrane potential of permeabilized fibers with the O2k-TPP+ ISE-Module required adaptations in the experimental handling, especially the introduction of the sample into the already calibrated chamber. A more specific problem for this group was how to achieve re-oxygenation when the use of the catalase / H2O2 method is not possible and the usual method of opening the chamber would again disturb the already attained calibration of the TPP electrode. Solutions for both problems were initially described by "Peter" Lin Chien-Te from the Neufer group on this Discussion page and later incorporated into Mitochondrial_membrane_potential#Mitochondrial_membrane_potential_of_permeabilized_fibres.
