Property "Has-20abstract" (as page type) with input value "-5B-5BImage:MITOEAGLE-2Dlogo.jpg-7Cleft-7C100px-7Clink=http:-2F-2Fwww.mitoeagle.org-2Findex.php-2FMitoEAGLE-7CCOST-20Action-20MitoEAGLE-5D-5D-0AAlthough-20the-20alternative-20oxidase-20(AOX)-20is-20naturally-20absent-20in-20the-20mitochondria-20of-20vertebrates-20and-20insects,-20its-20xenotopic-20expression-20in-20model-20organisms-20can-20attenuate-20spanerse-20phenotypes-20related-20to-20mitochondrial-20diseases,-20as-20this-20creates-20an-20extra-20path-20for-20oxygen-20reduction-20when-20the-20cytochrome-20c-20segment-20of-20the-20electron-20transfer-20system-20(ETS)-20is-20overloaded.-20Due-20to-20its-20non-2Dproton-2Dpumping-20nature,-20AOX-20activity-20in-20plants-20is-20clearly-20associated-20with-20thermogenesis-20-5B1-5D.-20We-20have-20recently-20reported-20that-20AOX-2Dexpressing-20flies-20develop-20faster-20and-20have-20higher-20viability-20than-20control-20flies-20at-20low-20temperatures-20-5B2-5D,-20indicating-20that-20AOX-20is-20also-20thermogenic-20in-20-27-27Drosophila-27-27-20and-2For-20it-20can-20release-20developmental-20constraints-20imposed-20on-20the-20ETS.-0A-0ATo-20test-20which-20of-20these-20mechanisms-20play-20a-20role-20in-20this-20AOX-2Dinduced-20cold-20advantage-20during-20development,-20we-20measured-20mitochondrial-20oxygen-20consumption-20on-20larval-20homogenates-20of-20the-20wild-2Dtype-20w-3Csup-3E1118-3C-2Fsup-3E-20and-20the-203X-27-27tubAOX-27-27-20line-20-5B3-5D,-20at-2012-20and-2025°C.-20Surprisingly,-20no-20differences-20in-20coupled-20respiration-20and-20state-203-2Fstate-204-20ratio-20were-20detected-20when-20substrates-20to-20the-20two-20main-20dehydrogenases-20in-20flies,-20complex-20I-20(CI)-20and-20mitochondrial-20glycerol-2D3-2Dphosphate-20dehydrogenase-20(mGPDH),-20were-20used-20concomitantly.-20In-20addition,-20AOX-20inhibition-20with-20n-2Dpropyl-2Dgallate-20did-20not-20affect-20respiration-20in-20these-20conditions.-20However,-20when-20measured-20separately,-20CI-2D-20and-20mGPDH-2Ddriven-20coupled-20respiration-20at-2012-20and-2025°C-20spanerged-20in-20AOX-20flies,-20the-20former-20being-20~48-25-20higher-20and-20the-20latter-20~33-25-20lower-20than-20in-20w-3Csup-3E1118-3C-2Fsup-3E-20flies.-20At-2012°C,-20AOX-20inhibition-20led-20to-20a-20~37-25-20decrease-20in-20mGPDH-2Ddriven-20coupled-20respiration,-20and-20to-20a-20~30-25-20decrease-20in-20mGPDH-2Ddriven-20leak-20respiration.-0A-0AOur-20data-20suggests-20that-20AOX-2Dexpressing-20larvae-20are-20producing-20heat-20via-20mGPDH-2DAOX-20functional-20interactions,-20which-20uncouple-20mitochondria.-20To-20compensate-20for-20ATP-20not-20produced-20by-20the-20mGPDH-20action,-20these-20flies-20increase-20oxidative-20phosphorylation-20through-20the-20CI-20pathway.-20At-20low-20temperatures,-20this-20configuration-20becomes-20highly-20functional,-20especially-20because-20mGPDH-20appears-20naturally-20less-20sensitive-20to-20cold-20-5B4-5D.-20Since-20mGPDH-20directly-20connects-20cytosolic-20processes-20with-20ETS,-20we-20can-20predict-20that-20the-20cellular-20redox-20state-20of-20the-20larvae-20is-20altered-20in-20the-20presence-20of-20AOX,-20affecting-20other-20metabolic-20processes-20at-20low-20temperatures.-20Furthermore,-20higher-20CI-2Ddriven-20coupled-20respiration-20could-20be-20a-20result-20of-20an-20increase-20in-20the-20tricarboxylic-20acid-20cycle-20reactions,-20which-20in-20turn-20would-20accelerate-20larval-20growth-20via-20increased-20cataplerosis.-20In-20combination,-20our-20data-20supports-20mGPDH-2Dlinked-20thermogenesis-20and-20CI-2Dlinked-20accelerated-20cataplerosis-20as-20explanations-20for-20the-20increase-20in-20fitness-20observed-20for-20AOX-2Dexpressing-20flies-20at-20low-20temperatures." contains invalid characters or is incomplete and therefore can cause unexpected results during a query or annotation process.
