Abstract: Conspicuous alterations in mitochondrial morphology have been observed in this laboratory (Wollenberger and Schulze, 1961, 1963) in the left ventricular cardiac musculature of dogs with experimental aortic stenosis of 4 months to 2% years standing. The major change seen at the stage of compensatory hypertrophy of the muscle prior to the development of cardiac failure was a disappearance of cristae. The electron micrographs revealed, in addition, a relative decrease in total mitochondrial volume (Wollenberger and Schulze, 1962, 1963). In the present communication these findings are complemented with information on mitochondrial function. As will be seen from the data presented below, the loss in the inner structure of the mitochondria of the hypertrophied ventricles was paralleled by a decline in the oxidative and phosphorylative power of the isolated organelles. Taken in conjunction with the diminution of the mitochondrial cell compartment, this decline-provided it does not represent a purely in vitro phenomenon-may be interpreted as reflecting a rather serious encroachment on the metabolic reserve of chronically overloaded and hypertrophied myocardium, occurring in advance of overt circulatory failure.
