Are you planning to perform several measurements in one day using the same O2k-chamber?
To avoid any possible contamination by inhibitors in the subsequent measurements, the O2k-chamber needs to be cleaned each time with the standard protocol AfterUse or AfterUse_inhibitors.

Does your polarographic oxygen sensor (POS) need a deep clean?

Add catalase when measuring Complex IV (CIV) activity in permeabilized fibers (pfi)
Pfi have a high content of iron-containing proteins, which accelerate the autoxidation of tetramethyl-p-phenylenediamine (TMPD) in the CIV assay. Catalase prevents the accumulation of H2O2 generated by this autoxidation.

Do you observe simultaneous fluctuations in the O2 slope neg. and the Peltier power?
